As the biopharmaceutical industry transitions toward
Industry 4.0, the adoption of robust Process Analytical Technology (PAT) is
critical for accelerating development timelines and reducing the environmental
footprint of manufacturing. Managing the inherent structural complexity of
monoclonal antibodies (mAbs) and peptides requires analytical techniques that
move beyond traditional offline testing.
A-TEEM (Absorbance–Transmittance Excitation Emission Matrix)
spectroscopy, coupled with advanced chemometrics, provides a multidimensional
"fingerprint" that enables rapid protein characterization. This
technique delivers high-resolution data with minimal sample preparation,
offering a scalable solution for real-time monitoring and in-line, on-line, or
at-line integration.
We present a framework for utilizing A-TEEM in
protein identification, quantification, and stability analysis. Key focus areas
include its implementation for real-time stability tracking and its efficacy in
monitoring Critical Quality Attributes (CQAs). By integrating A-TEEM into the
manufacturing workflow, users can achieve enhanced process understanding and
ensure product consistency from R&D through to commercial production.
